I am attempting to determine what all needs replacement in the defrost system. The thermostat has 6.1k ohms at room temperature and I haven't read the heater yet. What reading should I get there and what other tests should I perform if the redings are in the good range?

The defrost element should be around 30 ohms.
The defrost thermostat should be 0 ohms when frozen. Unplug the unit and one side of the defrost thermostat then hold it's face not an ice cube to see if it will close.
The resistance you see is probably normal when it is open.
If the defrost thermostat is bulged/deformed replace it even if it does measure OK.

Try to force a defrost, see the tech sheet.
If the heater comes on then probably the control board needs replacing.
Be careful that you do not burn your fingers.
