My whirlpool fridge is not cooling but the freezer is freezing over. manually defrosted the unit. I turned it on and it started to cool. It went into defrost one time. Overnight it froze up again. I uplugged it for just thirty minutes, and the unit defrosted. I did some more research. One site said to manually advance the defrost timer until it clicks. I did this, and the unit went into defrost on it's own and came out of
defrost and started cooling again. Which part could be defective?

What is the model number of your refrigerator?

In general it has been my experience that if the defrost timer advances it's ok.
This would leave you with either a faulty defrost heater or defrost thermostat.

I would put my money on a faulty defrost thermostat.
Sometimes you can tell if the defrost thermostat is faulty by simply looking at it.
In this photo the one on the left side is bad, see how the back of it is popped out of place.

To check it with a multimeter a good defrost thermostat should check closed when it's in a cold freezer.
